dc.description | Las plantas y los microorganismos se encuentran entre los recursos naturales con importancia en aplicaciones biotecnológicas. Los estudios realizados en algunas especies de la familia Gesneriaceae demuestran que tienen actividades biológicas prometedoras como antioxidantes, anticancerígenos, antiinflamatorios. Por tanto, este trabajo tuvo como objetivo general aislar, caracterizar y conservar microorganismos endofíticos que interactúan con Seemannia sylvatica (Kunth) Hanst (Gesneriaceae) y establecimiento de cultivos celulares in vitro. Para el aislamiento de microorganismos endofíticos se procedió a emplear dos métodos: 1) se realizaron cortes en la hoja (lámina) eliminando los bordes y la nervadura central y 2) se maceraron 2 gramos de hojas con solución PBS, en ambos métodos se emplearon 13 medios de cultivos incubando a la misma temperatura por 7 días, finalizado el tiempo se realizó caracterización morfológica. Las cepas identificadas se conservaron por ultracongelamiento y por el método Castellani, a su vez se realizaron cultivos in vitro para la obtención de callos y plántulas. Se logró el aislamiento de microorganismos siendo un total de 72 cepas, correspondiendo a 34 cepas fúngicas y 38 cepas de levaduras y bacterias, fue posible realizar la descripción macroscópica a cada cepa aislada observando que las colonias presentaron morfologías totalmente diferentes, a su vez fue posible realizar la conservación a partir de los dos métodos empleados evidenciando que ambos son efectivos para la conservación a largo plazo. De igual manera se logró la obtención de plántulas a partir de organogénesis directa empleando las hormonas ANA y BAP a diferentes concentraciones, se debe resaltar que los cultivos in vitros obtenidos fueron sometidos a diferentes procesos de limpieza y desinfección del material vegetal evidenciando que el protocolo estandarizado en este estudio fue el más efectivo, estos cultivos presentaron crecimiento a partir de la semana 14 en condiciones ambientales controladas. El proceso de callogénesis no fue efectivo debido a que las concentraciones empleadas de las hormonas 2,4-D y BAP no fueron las acertadas y se dio la presencia de contaminación en todos los tratamientos. | es_CO |
dc.description.abstract | Plants and microorganisms are among the important natural resources in biotechnological applications. Studies conducted in some species of the Gesneriaceae family show that they have promising biological activities as antioxidants, anticancer, anti-inflammatory. Therefore, this work had as general objective to isolate, characterize and conserve endophytic microorganisms that interact with Seemannia sylvatica (Kunth) Hanst and establishment of in vitro cell cultures. For the isolation of endophytic microorganisms, there were use two methods: 1) cuts were made in the leaf (sheet), eliminating the edges and midrib, and 2) 2 grams of leaves were macerated with PBS solution, in both methods were used 13 crop media incubating at the same temperature for 7 days, after the time, morphological characterization was performed. The identified strains were preserved by deep-freezing and by the Castellani method, at the same time in vitro cultures were performed to obtain callus and seedlings. Isolation of microorganisms was achieved, being a total of 72 strains, corresponding to 34 fungal strains and 38 strains of yeast and bacteria, it was possible to perform the macroscopic description of each isolated strain observing that the colonies presented totally different morphologies, simultaneously it was possible to do conservation with the two methods used, showing that both methods are effective for the conservation in a long-term. Likewise, it was possible to obtain seedlings from direct organogenesis using the hormones NAA and BAP at different concentrations, it should be noted that the in vitro cultures obtained were subjected to different cleaning and disinfection processes of the plant material, evidencing that the standardized protocol in this study was the most effective, these crops showed growth from week 14 under controlled environmental conditions.
