| Abstract: | Canine Monocytic Ehrlichiosis (CME) is a tick-borne disease affecting all canids, the etiologic agent is Ehrlichia canis, belonging to the Anaplasmataceae family, this multisystemic disease affects canines regardless of breed, sex or age, and is distributed worldwide, it is transmitted by the vector Rhipicephalus sanguineus, which belongs to the Ixodidae family found in warm climates. In this study, 51 blood samples corresponding to patients positive for CME by immunochromatography, aged between 1 month and 10 years of age, were selected. The DNA samples were extracted by the phenol-chloroform method and using a commercial kit. A PCR was developed for molecular diagnosis with the 16S rRNA gene, positive patients were subjected to an end-point PCR using the primers TRP36-F2 and TRP36-R1, then a hemi-nested PCR was used using the primers TRP36-F2 and TRP36-DF. The amplified samples were sequenced using the Sanger technique to determine variations in the trp36 gene in order to characterize and genotype E. canis by determining the tandem repeats. The analysis determined the presence of the United States (US) genogroup in 2 samples and the Costa Rican (CR) genogroup in 2 other samples. This work is the first to determine the genogroups of this disease of great importance in the canine population of the region due to its high prevalence. |
| Description: | La Ehrlichiosis Monocítica Canina (EMC) es una enfermedad transmitida por garrapatas que afecta a todos los cánidos, el agente etiológico es Ehrlichia canis, perteneciente a la familia Anaplasmataceae, esta enfermedad multisistémica afecta a los caninos sin distinción de raza, sexo o edad, siendo de distribución mundial, es transmitida por el vector Rhipicephalus sanguineus, que pertenece a la familia Ixodidae que se encuentra en climas cálidos. En este estudio, se seleccionaron 51 muestras de sangre correspondientes a pacientes positivos a EMC por inmunocromatografía, con edades comprendidas entre 1 mes y 10 años de vida, estas muestras fueron sometidas a extracción de ADN usando el método fenol- cloroformo y utilizando un kit comercial. Se desarrolló una PCR para el diagnóstico molecular con el gen 16S rRNA, los pacientes positivos, se sometieron a PCR punto final utilizando los primers TRP36-F2 y TRP36-R1, posteriormente, se utilizó una PCR semi-anidada utilizando los primers TRP36-F2 y TRP36-DF. Los amplificados fueron secuenciados mediante la técnica de Sanger para determinar variaciones en el gen trp36 con el fin de caracterizar y genotipificar E. canis determinando las repeticiones en tándem. El análisis realizado determinó la presencia del genogrupo Estados Unidos (US) en 2 muestras y el genogrupo Costa Rica (CR) DETERMINACIÓN DE GENOGRUPOS DE Ehrlichia canis 14 en otras 2 muestras. Este trabajo es el primero que se realiza determinando los genogrupos de esta enfermedad de gran importancia en la población canina de la región debido a su alta prevalencia. |
